Rational Genome To Proteome Prospecting™
There has been great progress in uncovering gene expression patterns that are clinically informative, and suggest pathway associations with disease. Nevertheless, key challenges remain to translate these gene relationships to new diagnostics and ultimately to new therapies. The foremost challenge is the poor correlation between gene expression data and Mass Spec proteomic data.

 

ProFACT's services assist gene expression researchers to overcome these challenges by providing:

  • A bioinformatics bridge to determine the most practical measurable protein attributes from a pre-selected gene panel,
  • Rational Proteome Prospecting to produce enriched gene products for final identification by the clent.

Starting from an initial client-selected gene panel and protein annotation from that panel, we coordinate disparate bioinformatics to index the gene's predicted tryptic peptide products and their suitability and specificity for polyclonal antibody production. Along with functional measurements (if available), such antibodies will score the presence of the full length gene product in each of the SeraFILE -derived subfractions as part of an enrichment strategy; see the link Rational Proteome Prospecting. Once the bioinformatics component is completed, consultation with the client considers the scope and cost of the project, and produces a final assignment of gene priorities, and injection of success milestones to continue. Discussions surrounding sample types, lysis conditions, and Mass Spec training can build confidence in the final Mass Spec reconciliation and verification to the selected gene targets. Once gene products are verified, SeraFILE can optionally be adapted for generating functional molecular profiles and biomarker assays.
